Manipulating interferon levels toward a safe and effective live single-cycle pediatric RSV vaccine

SUMMARY RSV is the largest viral cause of pediatric bronchiolitis and pneumonia, causing >100,000 deaths in children worldwide. Recently, the FDA approved a maternal vaccine to protect newborns. This vaccine is given to pregnant women to protect infants in the first months of life via passively acquired antibodies. This modality is a momentous step forward in the battle against RSV. However, as maternal antibodies wane, infants > 4-6 months remain vulnerable. In addition, not all may elect a vaccine in the third trimester; For this and other reasons, the young infant population will likely continue to present a range of protective Ab levels. Moreover, the burden of RSV disease and cost is high for children up to 4-5 years of age, and a vaccine to directly induce immunity in infants and children remains a priority. Intranasally applied live vaccines are an attractive approach as they induce a response that is broad (humoral, cellular, multi-antigenic i.e. not dependent on a single antigen) and includes mucosal immunity, which provides protection against upper respiratory tract infection and virus spread in the population. The primary aim of this proposal is to generate and test a novel live-attenuated vaccine for neonatal infants (< 28 days). However, live vaccines, especially in neonates, have unique safety/efficacy challenges. To prevent live vaccine reversion to virulence, we previously developed intranasal single-cycle RSV vaccines (see 'Significance'). Single-cycle vaccines are live and produce high levels of viral antigens, but are blocked in cell-cell transmission and cannot revert to virulence, including in context of suboptimal immunity. In adult mice, prime-boost vaccination with single-cycle vaccine induced broad, robust, immunity and protected mice from challenge. Another challenge for live neonatal vaccines is immunologic immaturity. Whereas this may explain generally weak neonatal responses, some studies suggest that even young infants have potential to induce high Ab titers. Immaturity can however lead to unbalanced Thelper cell responses (Th2-bias) and related immunopathology after re-exposure to RSV, reminiscent of a failed trial in the 1960s using inactivated RSV vaccine. A main factor believed to underlie neonatal Th2 bias is a weak early type I interferon (IFN) response, as addition of exogenous IFN alleviated the bias. For an effective vaccine, we propose to improve the IFN response through distinct mechanisms (blocking viral IFN antagonism, and integration of IFN expressing genes) and to test the impact in a neonatal mouse model. In addition, our vaccine design focuses the fusion and attachment protein responses for improved cross-efficacy, and it's intranasal route of vaccination should reduce inhibition by maternal antibodies, which we will investigate. We expect the combined enhancements to deliver a prime-boost vaccine that is both safe (no Th2 bias) and efficacious (protective against multiple strains) with a prime vaccination given at a neonatal age. Finally, we also investigate neonatal versus post-neonatal prime-vaccination. This broadens the impact of our study to include potential for alternatives, such as a post-neonatal vaccination modality (prime at 1-4 months) or a modality to work in conjunction with the Pfizer maternal vaccine (prime at 4-6 months).

Manipulating PIWI-interacting RNA to generate sterile ticks

SUMMARY The Piwi-interacting RNA (piRNA) pathway is a conserved genome surveillance mechanism essential for germline integrity and fertility in invertebrates, including ticks. Piwi (P-element Induced Wimpy Testis) proteins play a critical role in silencing transposable elements (TEs) to preserve genome stability in germ cells. Our preliminary data shows that depletion of Piwi transcript in ticks leads to reproductive failure, establishing a novel target for developing sterile insect technology (SIT) against ticks. The overall goal of this application is to develop a mechanistic framework for piRNA-based SIT by manipulating the tick piRNA pathway and evaluating its impact on fertility and pathogen transmission. Our central hypothesis is that disrupting the piRNA pathway in ticks will induce sterility by derepressing transposable elements, impairing germline genome integrity, and ultimately reducing vector fitness. To test this hypothesis, we propose a single aim to disrupt the piRNA pathway in ticks to induce sterility and reduce vector competence. This study will define a Piwi knockdown signature characterized by TE derepression and sterility. These experimental findings will establish a rational foundation for the development of a piRNA-based SIT platform targeting tick vectors. Moreover, the outcome of this project could be extended to other tick species of public health significance.

Manipulation of host cell peroxisomes by the bacterial pathogen Legionella

PROJECT SUMMARY Peroxisomes are essential organelles that play pivotal roles in diverse biochemical pathways, from lipid metabolism and homeostasis to reactive oxygen detoxification. Peroxisome activities are intimately linked with other organelles, both functionally, coordinating metabolic pathways, and physically, through membrane contact sites that allow direct transport of metabolites between sub-cellular compartments. Recently, peroxisomes have been recognized for acting as signaling platforms, with key roles in immunity. Thus, a pathogen’s ability to modulate peroxisome biology could have profound impacts on numerous host cellular processes. However, the role of peroxisomes in bacterial pathogenesis remains largely unexplored. Recent findings indicate that bacterial pathogens exploit peroxisome biology to subvert host defenses. Many intracellular bacterial pathogens reside within membrane-bound compartments, shielding the bacteria against recognition and killing by host immune surveillance pathways. Maintaining the integrity of these replication vacuoles is critical for pathogen survival and virulence. Recently, we discovered an unprecedented link between bacterial pathogens and peroxisomes in which mutations that render peroxisomes metabolically inactive restrict replication vacuole expansion, induce vacuole rupture and limit bacterial replication. In the case of Legionella, we found that this pathogen actively manipulates host peroxisomes, altering the relative abundance of peroxisome metabolic enzymes, and recruiting peroxisomes to its replication vacuole. These processes are dependent on the bacterium’s Type IV secretion system, implicating Legionella secreted effector proteins in peroxisome modulation. However, the effectors involved remain unknown. The goal of this proposal is to elucidate how Legionella manipulates host peroxisome biology and to define the role of peroxisomes in Legionella pathogenesis. We will identify and characterize the bacterial effectors responsible for altering peroxisome metabolic programming and location, providing critical insight into the virulence strategies employed by Legionella. By uncovering novel effector functions and host targets, this work will advance the molecular understanding of bacterial pathogenesis and host cell manipulation. Moreover, this research will address a critical gap in our understanding of how bacterial pathogens exploit host organelles to establish infection. Given the emerging evidence that other intracellular bacterial pathogens - Salmonella, Mycobacterium and Chlamydia – also exploit peroxisomes, our studies are expected to reveal broadly relevant virulence strategies.

Manipulation of leucine uptake, sensing, and metabolism in macrophages as a diagnostic and therapeutic strategy in atherosclerosis

Project Summary / Abstract Despite numerous improvements in treatment and prevention, cardiovascular disease (CVD) remains one of the leading causes of morbidity and mortality worldwide. Nutrients such as lipid, particularly derived from the diet, have long been linked to the development and exacerbation of CVD. However, the role of other nutrients such as amino acids in the pathogenesis of CVD has largely remained unexplored. In work spanning the past decade, we have implicated amino acids as key stimuli in pathogenic mTOR signaling in macrophages via a lysosomal nutrient-sensing mechanism. We further identified leucine as the single most important nutrient driving this lysosomal mTOR signaling in atherosclerosis. A significant contributing factor to leucine-mediated signaling is the concomitant increase in a major leucine transporter, SLC7a5/LAT1. Macrophage Slc7a5-deficiency significantly reduces mTOR signaling and plaque size and complexity in atherogenic mice. We have also leveraged the upregulation of leucine transport as a diagnostic approach to imaging atherosclerotic plaques by demonstrating the affinity of a leucine-like PET radiotracer to macrophage-rich areas of lesions. Beyond leucine uptake, we recently discovered that its catabolism is inhibited in plaque macrophages which exacerbates leucine accumulation and mTOR. Overall, we provide multiple promising approaches to modulating leucine-mTOR signaling in macrophages. In this proposal, we will impinge on each of these processes by genetic and pharmacological means in order to test the hypothesis that perturbation of leucine uptake, lysosomal sensing, and metabolism can be effective diagnostic and therapeutic strategies in atherosclerosis. This will be achieved in three distinct aims: 1) to evaluate the diagnostic and therapeutic potential of modulating macrophage leucine uptake in atherosclerosis, 2) to dissect the salient leucine-sensing mechanisms at the lysosome in driving mTOR activation and atherosclerosis, and 3) to evaluate the therapeutic potential of enhancing macrophage leucine catabolism in atherosclerosis. We will use an innovative set of tools including novel PET probes and a diverse array of mouse models to pursue a highly mechanistic evaluation of macrophage leucine-mTOR signaling in CVD.

Manipulation of the systemic environment to improve skeletal muscle recovery after disuse-induced atrophy

PROJECT SUMMARY / ABSTRACT Aging Veterans have higher incidences of comorbid chronic diseases compared to the general population, resulting in more frequent hospitalizations. An outcome of frequent hospitalizations in aged individuals is prolonged bed rest or immobilization, which accelerates sarcopenia. This period of disuse is compounded by aging, as skeletal muscle mass and function fail to fully recover to the same extent as in younger adults. The failure to recover skeletal muscle mass and function after disuse atrophy has a profound impact on mobility, independence, and quality of life. Therefore, improving the ability of aged skeletal muscle to recover muscle mass and function after periods of disuse would improve clinical outcomes in the lives of aging Veterans. Exercise is polytherapeutic in that it positively affects most tissues and attenuates many of the hallmarks of aging. Exercise releases factors such as peptides, metabolites, and RNAs into the systemic environment (termed exerkines), many of which are transported via extracellular vesicles (EVs). These exerkines act in an autocrine, paracrine, and endocrine manner to improve health outcomes. The current study tests the novel idea that changing the systemic milieu with exercised plasma and exerkine-packaged EVs isolated from plasma will improve skeletal muscle health after disuse atrophy. These data will be the first to explore how the systemic environment could be manipulated to improve muscle recovery after bed rest using exercised plasma and EVs. The overall hypothesis is that exerkines in plasma and EVs can improve the recovery of aged skeletal muscle mass and function after a period of disuse-induced atrophy. To test this hypothesis, we will administer exercised plasma before, during, or after hindlimb unloading (HLU) to determine the effects on skeletal muscle recovery in both sexes. We will also determine the effects of EV-depleted plasma, isolated exercise-induced EVs, and synthetic “exercise-mimicking” EVs on aged skeletal muscle recovery and mitochondrial structure and function after a period of HLU. We will use a battery of approaches to assess functional and molecular changes in skeletal muscle mass and function, as well as how each intervention alters the systemic environment. We will examine plasma and EV cargo using proteomic and miRNA sequencing to determine potential mechanisms of recovery. To delineate potential mechanisms, we will use Support Vector Machine Learning to determine which miRNAs and proteins independently and concomitantly improve aged skeletal muscle recovery. We expect improved muscle recovery following the administration of exerkines in plasma and EVs (endogenous and synthetic EVs), and that these improvements will be associated with increased mitochondrial bioenergetic capacity, redox balance, and anti-inflammatory processes. Furthermore, we will identify which miRNAs and proteins are responsible for these improvements in skeletal muscle mass and function. My short-term goals are to determine if manipulation of the systemic environment with exercise plasma and plasma factors can improve aged skeletal muscle mass and function after disuse atrophy. To achieve these short-term goals, I will receive training in aging, the use of stable isotopes, EVs, bioinformatics, and scientific communication. I will receive this training through a combination of meetings with my mentoring team, conferences, presentations, formal courses, and conducting experiments. The training plan I have outlined will allow me to achieve my long-term goal of becoming an independent VA scientist.

The National Institute of Standards and Technology (NIST) invites proposals from qualified organizations for projects in the areas noted below. These activities will enhance the Manufacturing Extension Partnership's (MEP)relationships with state stakeholders and partners to improve productivity and technological performance in United States manufacturing, achieving the MEP mission through the participation of individuals from industry, universities, State governments, other Federal agencies. All organizations meeting eligibility requirements provided in the Federal Funding Opportunity (Full Announcement) are invited to submit proposals that address both of the key areas below: 1. Network building and expansion: The Recipient will collaborate with NIST MEP, MEP centers, and partners to facilitate the development of effective working relationships for MEP Federal and Center officials among a wide range of stakeholders. Stakeholders include state agency leaders, policy makers, practitioners involved in technology and workforce-based economic development planning and program implementation, and others as suggested by the Recipient. The Recipient, the MEP system, and NIST MEP will share information about the needs and agendas of state stakeholders, risks and opportunities, using their in-depth understanding of budget data, current trends, emerging issues. The Recipient will propose ideas that help to implement initiatives for encouraging a dialogue among interested parties, including states, MEP Centers, partner organizations, and industry, about how the MEP partner network and MEP next generation initiatives best fit into state strategies and priorities, and how manufacturers and the public benefit from manufacturing extension activities. The Recipient will work with entities in the MEP system to help prioritize states for attention, develop strategies to address opportunities and concerns, recommend effective communication strategies, and convene meetings of the MEP Centers, partners and stakeholders as needed to facilitate the partnership development. The Recipient will demonstrate the ability to provide nationwide access to state relations support services. 2. Research/knowledge development and sharing: The Recipient will collaborate with NIST MEP, MEP centers, and partners to compile and analyze information available about current state budgets, economic development and technology policy initiatives, program evaluation practices, and the nature of the partnership between states, MEP Centers, industry and NIST. To help the partners in the MEP system sustain and improve state relationships, the Recipient will collaborate with the MEP system to track state activities, share insights and available research, provide technical assistance to resolve problems that arise, and undertake related activities to facilitate the desired partnerships. NIST does not prescribe any particular management or operational structure for this effort, but work proposed should be based upon approaches or structures used by collaborative providers of support services that have demonstrated success.
