SUMMARY The Piwi-interacting RNA (piRNA) pathway is a conserved genome surveillance mechanism essential for germline integrity and fertility in invertebrates, including ticks. Piwi (P-element Induced Wimpy Testis) proteins play a critical role in silencing transposable elements (TEs) to preserve genome stability in germ cells. Our preliminary data shows that depletion of Piwi transcript in ticks leads to reproductive failure, establishing a novel target for developing sterile insect technology (SIT) against ticks. The overall goal of this application is to develop a mechanistic framework for piRNA-based SIT by manipulating the tick piRNA pathway and evaluating its impact on fertility and pathogen transmission. Our central hypothesis is that disrupting the piRNA pathway in ticks will induce sterility by derepressing transposable elements, impairing germline genome integrity, and ultimately reducing vector fitness. To test this hypothesis, we propose a single aim to disrupt the piRNA pathway in ticks to induce sterility and reduce vector competence. This study will define a Piwi knockdown signature characterized by TE derepression and sterility. These experimental findings will establish a rational foundation for the development of a piRNA-based SIT platform targeting tick vectors. Moreover, the outcome of this project could be extended to other tick species of public health significance.
