Hippo and Wnt pathway regulation of mammalian Muller glial cell reprogramming and proliferation

NEI - National Eye Institute

Retinal diseases such as glaucoma, macular degeneration, and diabetic retinopathy, as well as traumatic injury, result in loss of retinal neurons and thus sight, depriving many worldwide of one of our most valued senses. Thus, there is a critical need to devise strategies to restore lost retinal neurons, leading to vision recovery. Current efforts in retinal regenerative medicine are heavily invested in cell replacement approaches. However, it may also be possible to induce the mammalian retinae to undergo an intrinsic self-repair mechanism to regenerate neurons. The retinae of non-mammalian vertebrates, such as zebrafish, are known to exhibit the remarkable ability of retinal regeneration. Here, Müller glial cells (MGs) reprogram to proliferative, retinal progenitor-like cells that, in turn, differentiate into new photoreceptors, leading to restoration of vision. Unfortunately, for unknown reasons, mammalian MGs have lost this ability, or it is dormant. Our long-term goal is to identify the cellular and molecular mechanisms blocking mammalian MG-mediated retinal regeneration. By doing so, we may be able to devise strategies to bypass this system and thereby reawaken the regenerative potential of the mammalian retina. In this proposal, we will test the overarching hypothesis that transient modulation of the Hippo signaling pathway drives MGs into a progenitor-like state capable of generating new retinal neurons and MGs. To test this hypothesis, we will employ a multi-disciplinary approach using genetic loss- and gain-of-function experiments, fate mapping, and single-cell transcriptomics. Our specific aims will precisely determine the fate of proliferative MGs, which are non-responsive to Hippo pathway regulation, as being neurons and/or MGs. We will also determine how retinal damage triggers MG cell cycle entry prior to repression by Hippo. These data will provide an essential molecular entry point for further investigating novel methods to promote Müller glial cell-mediated retinal regeneration, and are likely to have a significant impact on the field of retinal regenerative medicine. We anticipate subsequent investigation into more translational, therapeutic methods to modulate Hippo pathway activity to promote retinal regeneration.

Hispanic Serving Institutions: Equitable Transformation in STEM Education (ETSE)

Hispanic Serving Institutions (HSI) are an important component of the nation s higher education ecosystem and play a critical role in realizing the National Science Board Vision Report for a more diverse and capable science and engineering workforce. Aligned with this vision and the NSF Strategic Plan 2022 -2026 the goals of the NSF HSI Program are to: 1. Enhance the quality of undergraduate science, technology, engineering, and mathematics (STEM) education at HSIs. 2. Increase the recruitment, retention, and graduation rates of students pursuing associate s or baccalaureate degrees in STEM at HSIs. Meeting these goals requires institutions to understand and embrace their students strengths, challenges, identities and lived experiences. This can happen in many ways and across many areas of an institution. As such, the IUSE: HSI program provides multiple opportunities to support an institution s goal to become more student centered, including theEquitable Transformation in STEM Education (ETSE) competition. This competition includes the following tracks: Departmental/Division Transformation Track (DDTT) - New Institutional Transformation Track (ITT) Emerging Faculty Research Track (EFRT) - New HSI Program Resource Hubs (Hubs) This solicitation will also accept conference proposals and planning proposals, as defined by the PAPPG. The ETSE competition focuses on (1) institutional transformation projects that support HSIs in their effort to achieve equity in STEM education, and (2) the infrastructure the HSI-Net network of resource hubs which supports the overall program goals. Institutions are encouraged to consider how their HSI designation, and their organizational mission align to better support STEM success of all students. The ETSE competition welcomes proposals that look to implement and evaluate promising practices and/or conduct research related to broadening participation or improving recruitment, retention, graduation, and other successful outcomes in STEM undergraduate education. The ETSE solicitation supports projects designed to catalyze change and help HSIs meet students where they are, accounting for their assets and the challenges they may face. Identities and experiences are not determined solely by membership in a single monolithic population of students (e.g., Hispanic, first-generation, commuter, etc.). Consequently, institutions are expected to use institutional data to identify equity gaps, identify areas of need, and unpack the factors that shape students individual identities and shared experiences. The perspectives gained from this data should be central to the design of the proposed project. Please see below for specific information about each track. While proposals are focused on mechanisms for transforming undergraduate STEM education, projects should also consider student voices and include mechanisms to aggregate and analyze existing student feedback and collect quantitative and qualitative student data throughout the life of the proposed project.

Histone methyltransferase, Setd2, in substance use disorders

Substance use disorders (SUDs) are characterized by long-lasting neuroadaptations, including changes in gene expression and the epigenome. The relevance of the epigenome in SUDs has been shown using overexpression or inhibition of epigenome modifying enzymes. However, such manipulations cannot distinguish between direct and indirect action of epigenetic modifications at a target gene. To address this, we apply locus-specific epigenetic editing with CRISPR/dCas9 to establish the direct causal relevance of a specific epigenetic modification to SUDs. We and others find that the epigenetic enzyme, SET domain containing 2 (Setd2), regulates cocaine-induced mouse behavior, gene expression, and alternative splicing. Setd2 specifically writes H3K36me3, a histone modification associated with active transcription and alternative splicing, which is altered in mouse brain regions by cocaine exposure. We and others have identified a role for Setd2/H3K36me3 in alternative splicing which mirrors cocaine-induced splicing events and is associated with cocaine-related behaviors in mice. We find that overexpression of Set2 in mouse brain reward regions increases H3K36me3 and drives mouse cocaine reward behavior. Together, these data provide strong evidence for Setd2/H3K36me3 in mediating the behavioral response to cocaine in mice through epigenetic and alternative splicing events. The overall goal of this proposal is to establish that Setd2 inhibition attenuates cocaine self-administration through cell-type specific regulation of alternative splicing and gene expression. First, we will determine both sufficiency and necessity of Setd2 to drive cocaine behavior, using pathway-specific Setd2 overexpression and small-molecule Setd2 inhibition (Setd2i), respectively. We hypothesize that NAc Setd2 inhibition will reduce cocaine self-administration. Second, we will define the pathway-specific effects of Setd2 on behavior, H3K36me3 and gene expression by cell-type specific epigenomic profiling and targeted epigenetic editing. Third, we will interrogate mechanisms by which Setd2 regulates alternative splicing by identifying interacting RNA-binding proteins (RBPs). Finally, this proposal will investigate non-enzymatic roles of a histone modifying enzyme in addiction. Recent studies and our preliminary data suggest non-enzymatic function of Setd2 through protein binding domains. We will distinguish Setd2 and H3K36me3 using combinatorial Setd2 overexpression, Setd2i, and epigenetic editing.

Historic Resource Study of Early Civil Rights Movement in National Capital Region
