The NIST RET will allow the selected teachers to participate in scientific research with NIST scientists and engineers at the NIST Campus in Gaithersburg, Maryland, that will encourage the teachers to inspire their students to pursue careers in fields of Science, Technology, Engineering, and Mathematics (STEM). The selected teachers will be required to attend the NIST RET during the summer of 2011 (exact dates to be decided by the teachers and scientists).

NIST Summer Institute for Middle School Science Teachers

The National Institute of Standards and Technology (NIST) is soliciting applications from qualified public school districts or accredited private educational institutions that are teaching students in the areas of Science, Technology, Engineering and Mathematics (STEM) at the middle school level (Grades 6-8). Such schools may offer instruction in general science fields including earth science, physical science, chemistry, physics, and/or biology. NIST will award funding that will support the attendance of middle school teachers in the NIST Summer Institute for Middle School Science Teachers (NIST SI), to be held July 6-19, 2010 to be held at the NIST Gaithersburg, Maryland, campus. The aim of the NIST SI is to increase teachers understanding of the subjects they teach, provide materials and resources to implement what they have learned at NIST in the classroom, enhance their enthusiasm for science, and provide them the opportunity to develop an on-going network with the scientists and engineers at NIST, who would be available for consultation even after the NIST SI has ended. The NIST Summer Institute is a hands-on workshop where middle school science teachers are able to take advantage of resources from the nation s standards and measurement laboratory, the National Institute of Standards and Technology. The workshop will provide teachers with instructional materials and ideas to use in their teaching, and will emphasize the measurement science done at NIST. The aim is to take a selection of the cutting-edge research at NIST and help the teachers translate this through hands-on activities into classroom-based learning modules. In addition to ideas and activities to incorporate into lesson plans, teaching tools are directly provided to allow the teachers to demonstrate to their students the activities that they select from this workshop. The program will provide a world-class opportunity for those teaching our nation's next generation of scientists to learn more about the subjects they teach and the research in those subjects at NIST, and offer a platform from which teachers can inspire their students to pursue STEM careers. Teachers representing successful applicants will be supported via a small stipend, awarded through their respective school district or private educational institution.

Nitric Oxide Regulation in Alcohol Tolerance

PROJECT ABSTRACT Chronic alcohol consumption leads to pharmacological tolerance of alcohol, promoting excessive alcohol-seeking behavior and resulting in alcohol use disorder (AUD). As of 2018, approximately 5.8% of the US adult population, or 14.4 million adults, suffer from AUD. Therefore, identifying the neurobiological mechanisms underlying alcohol tolerance and developing targeted treatments is crucial. Neurogranin (Ng) is expressed in the post-synapse of glutamatergic neurons and modulates NMDAR- mediated synaptic plasticity and nitric oxide (NO) bioavailability in the medial prefrontal cortex (mPFC). Ng null mice develop alcohol tolerance faster and self-administer more alcohol than wild-type mice following chronic alcohol exposure. Since NMDAR-mediated neuroadaptation occurs in the mPFC to compensate for the effects of alcohol, we hypothesize that Ng levels in the mPFC play a crucial role in NMDAR-mediated NO regulation, which maintains alcohol tolerance development. To address this mechanistic question, we will utilize a 16-hour operant conditioning model to study the high rate of alcohol self-administration and test whether mPFC-specific Ng knockout and overexpression regulate alcohol tolerance and alcohol seeking. We will then elucidate the impact of chronic alcohol on the Ng- NO pathway and its contribution to neuroadaptation in the mPFC using microdialysis and label-free proteomics. Aim 1 will investigate the molecular mechanism by which alcohol reduces Ng expression in the mouse mPFC. Subsequently, we will evaluate the development of alcohol tolerance using rodent behavioral tests following chronic binge alcohol self-administration in mPFC-specific Ng knockout and mPFC-specific Ng overexpression mouse models. Aim 2 will examine whether inducible Ng knockout mice in excitatory neurons (Cre-CaMKII-ERT2-Ng f/f) exhibit increased alcohol tolerance. Then, we will evaluate the effects of chronic alcohol consumption on glutamate-mediated NO neurochemistry using in vivo microdialysis and label-free proteomics. Aim 3 will investigate whether an NO donor (DetaNONOate) in the mPFC can prevent alcohol tolerance development and alcohol seeking in nNOS knock-out mice and glutamatergic neuron-specific Ng knock-out mice. By employing advanced molecular techniques and innovative behavioral assays, our study promises to advance the field significantly and provide a deeper understanding of alcohol's effects on mPFC function. This research addresses a critical gap in understanding alcohol's impact on NMDAR and mPFC. It also provides a novel mechanistic insight focused on the Ng-mediated NO pathway in the mPFC and its role in preventing alcohol tolerance. The outcome of the pharmacological treatment will provide new evidence of how maintaining NO levels in the mPFC precludes the development of alcohol tolerance and suggest potential therapeutic interventions for AUD.

Nitrogenase - From mechanistic investigation to heterologous expression

PROJECT SUMMARY Nitrogenase is a complex metalloenzyme central to the global nitrogen cycle, catalyzing the reduction of atmospheric nitrogen to bioavailable ammonia. Termed biological nitrogen fixation, this fundamental process provides ~200 million tons of reduced nitrogen annually, sustaining Earth's ecosystems. Despite decades of research, the (1) enzymatic mechanism and (2) heterologous expression of nitrogenase remain elusive, presenting opportunities for innovation. With respect to 1, the traditional mechanistic thinking of nitrogenase has been driven by the presumption of a structurally ‘static’ cofactor during catalysis. Our recent structural observation of asymmetric belt-sulfur displacements with distinct dinitrogen species in the two cofactors of Mo-nitrogenase invokes a novel mechanism of N2 reduction that necessitates dynamic structural rearrangements of the cofactors during catalysis. In the next 5 years, we will use genetic, biochemical, spectroscopic and structural approaches to probe this novel reaction mechanism, demonstrating the stepwise N2 reduction at the three belt-sulfur sites of each cofactor and illustrating the alternate interaction between the two components of Mo-nitrogenase that drives the coordinated reaction sequences via asynchronous rotation of the two cofactors. These studies will establish a much-needed framework for further exploration of the intricate reaction mechanism of nitrogenase and, in the long run, facilitate the development of nitrogenase-based applications. With respect to 2, efforts to heterologous express nitrogenase have been focused on introducing a complete gene set into a foreign host like E. coli via a ‘top-down’ approach. However, this approach faces challenges in ensuring proper assembly of the key intermediates that eventually give rise to a functional nitrogenase. Our recently developed ‘bottom-up’ strategy allowed a systematic reconstruction of the nitrogenase assembly pathway in E. coli and the ultimate expression of an active prototype nitrogenase, albeit with reduced activity under anaerobic conditions. In the next 5 years, we will use protein-engineering and synthetic-biology methods to enhance the functionality of our prototype nitrogen-fixing system through optimization of the nitrogenase assembly pathway, engineering of the electron and energy supplies in the E. coli host, and implementation of the mechanism of oxygen tolerance in this expression system. These efforts will provide strategies that can also be applied to the heterologous expression of other complex metalloenzymes, thereby advancing both fundamental science and general biotechnological innovation.

Nitrogenase - Where Haber-Bosch meets Fischer-Tropsch

PROJECT SUMMARY Nitrogenase catalyzes the reduction of atmospheric dinitrogen to bioavailable ammonia, a process that supports the existence of the entire human population. Additionally, nitrogenase reduces carbon monoxide to hydrocarbons, highlighting its potential for coupling environmental health with sustainable energy production. Unlike industrial Haber-Bosch (ammonia synthesis) and Fischer-Tropsch (carbon fuel synthesis) processes, nitrogenase operates at ambient conditions with protons and electrons as reducing agents, making it an attractive model for chemical energy conversion. Our long-term vision is to advance the mechanistic understanding of nitrogenase assembly and reactivity while leveraging its unique properties for innovative, environmental sustainability applications. In the next five years, we propose to use combined genetic, biochemical, spectroscopic and structural approaches to investigate how M-cluster, the unique metallocofactor of the molybdenum nitrogenase, is assembled into a functional unit, with a focus on the radical SAM-dependent carbide insertion concomitant the formation of an 8Fe cofactor core, the in vivo source and insertion mechanism of the ‘9th sulfur’. and the mobilization of Mo for cofactor maturation. Through our proposed studies, we expect to further refine the biosynthetic pathway of the unique metallocofactor of nitrogenase, which will provide crucial insights into the structural-functional relationship of this important enzyme and reveal general principles of the assembly mechanisms of complex metalloclusters in biological systems. Additionally, we will explore the reactivity of the vanadium nitrogenase, notable for surpassing its molybdenum counterpart in its capacity to reduce carbon monoxide to hydrocarbons. This unique trait of vanadium nitrogenase makes it a valuable tool for carbon recycling. In the next 5 years, we will use hybrid systems of vanadium nitrogenase to dissect its reactivity toward carbon monoxide by accumulating intermediates through modulation of electron flux (via heterometal variation), proton flux (via organic compound alteration), and electron availability (via mismatched electron donors). Through our proposed studies, we expect to identify novel strategies that can also be applied to the mechanistic investigations of enzymatic dinitrogen reduction, as well as establish prototype systems for developing nitrogenase-based applications that recycle carbon wastes into useful chemical products.

NJIA: A multi-level intervention to address stigma for women living with HIV in Tanzania

Achieving durable viral suppression among women living with HIV (WLWH) is critical to decreasing HIV transmission and ending the HIV epidemic in the United States and globally. However, for a subset of the population, sustained engagement in HIV services remains challenging. This situation is a primary contributor to persistent HIV transmission and a fundamental barrier to eradicating HIV. Stigma has been associated with suboptimal antiretroviral therapy adherence and viral load suppression. HIV stigma is a complex, multilevel barrier that has not been adequately addressed in existing service delivery. Although other barriers exist, stigma is a driving factor in low engagement of WLWH in HIV care, and evidence suggests that addressing stigma will improve sustained engagement in HIV care. Despite the well-documented negative impacts of stigma on HIV outcomes, there is a critical gap in innovative, evidence-based, multilevel interventions to reduce stigma. To address stigma and support lifetime HIV viral suppression among WLWH, we need multilevel strategies that can be readily implemented at scale. Our interdisciplinary and experienced team of Investigators will address this key gap. To support the multi-factorial research design required to rigorously evaluate multilevel stigma reduction interventions in a time- and cost-efficient manner, the following conditions are required: (1) A high geographical concentration of WLWH who struggle to engage with HIV care residing in the catchment area of a large enough number of HIV clinics to support randomization of health facilities and; (2) evidence-based stigma reduction interventions at both the health care facility (HCF) and individual WLWH levels to combine into a multilevel intervention. With all these conditions present, Tanzania offers a unique opportunity to evaluate the impact of a multilevel HIV stigma reduction intervention and generate translatable evidence for other settings where stigma is a significant barrier to sustained HIV care engagement but the conditions to evaluate a multilevel intervention are not present (e.g., the southern U.S.). This project builds on our team’s pioneering research on HIV stigma reduction interventions at the HCF and individual levels. We have previously adapted two complementary HIV stigma reduction interventions in Tanzania. The first, the Total Facility Approach (TFA), addresses HIV stigma among HCF staff. The second intervention, the Labda Siku Moja (LSM), is an internalized stigma reduction intervention. We will refine and combine these two interventions into a multilevel stigma reduction intervention to address HIV stigma faced by WLWH. In Aim 1, we will implement a hierarchical 2×2 factorial cluster-randomized controlled trial to determine the effectiveness of the multilevel stigma reduction intervention on quality of life and HIV viral suppression among WLWH who have missed scheduled clinic visits. In Aim 2, we will conduct a prospective mixed-methods process evaluation to assess implementation processes, theorized mediators influencing intervention effectiveness, and preliminary costs. In Aim 3, we will convene a series of study advisory group workshops to interpret Aims 1 and 2 findings and refine the intervention for broader implementation. The proposed research will fill a critical gap in our knowledge for supporting sustained engagement in lifetime HIV care, essential to eradicating HIV from the U.S. and globally. HCF and internalized stigma are significant barriers to maintaining viral load suppression and preventing onward transmission of HIV. This study provides the unique conditions necessary to evaluate a multilevel stigma reduction intervention and, if effective, will provide a translatable multilevel stigma reduction approach to fill the significant unmet need for stigma reduction in the U.S., thereby improving the lives of American WLWH and their families while advancing public health.
