Maternal prenatal stress and nutrition influence fetal growth and long-term child outcomes. Maternal perceived stress and undernutrition frequently co-occur and affect overlapping biological stress pathways related to oxidative stress, Still, little is known about the interactive effects of maternal prenatal stress and nutrition on fetal and long-term child outcomes and neurological development. The proposed R00 research will leverage data from the “Enhancing Nutrition and Antenatal Infection Treatment for Maternal and Child Health” (ENAT) trial to examine interactive effects of maternal prenatal stress and nutrition intervention on long-term child outcomes. The ENAT study randomized n=2390 pregnant women to receive an “enhanced nutrition” package or “standard care” and collected data on maternal prenatal stress (Cohen’s Perceived Stress Scale) throughout the pre- and postnatal period. Using data from ENAT, we will determine independent and interactive effects of maternal prenatal perceived stress and nutrition intervention on children’s cognitive and neural developmental at 24 months of age. Child outcomes will include stress-sensitive outcomes previously shown to be sensitive to early life adversity, including attention, memory, and language. We will also use electroencephalography (EEG) to examine neural oscillation across different frequency bands as an index of neural maturation. To study underlying biological pathways, we will use maternal and infant blood samples examine whether maternal and newborn telomere lengths may serve as biomarkers of in-utero programming of birth and child outcomes in relation to maternal prenatal perceived stress and nutrition. We hypothesize that higher levels of maternal prenatal stress will be associated with poorer cognitive and neural oscillatory child outcomes. Telomeres are non-coding tandem repeats at the end of the chromosomes that maintain genome and cell integrity. Telomeres erode over time and stress and poor nutrition can lead to accelerated erosion and cellular aging. Children with low birthweight have been found to have shorter telomeres. We therefore hypothesize that effects of prenatal stress on birth and child outcomes will be mediated by maternal and newborn telomere length, but that such associations will be attenuated in offspring of women who received nutrition intervention This R00 will enable me to lead an innovative research program in child neurodevelopment and establish me as a leader and innovative scholar utilizing advanced methods to uncover mechanistic processes that shape childhood development in domestic and global settings. The proposed research aligns with NICHD’s goal to set a foundation for healthy pregnancies and lifelong living. We will generate epidemiologic and biologic evidence linking maternal prenatal nutrition and stress with newborn and childhood outcomes that are sensitive to prenatal stress and form long-term outcomes related to school achievement and mental health. We will use this knowledge to guide public health decisions regarding pre- and postnatal intervention both domestically and globally, and to develop future intervention to support optimal child development and health.

Nutrient sensing in POMC neurons

Changes in metabolic states require the rapid and synergistic interaction between the brain and periphery to maintain homeostasis. Circulating factors such as nutrients play fundamental roles in these processes. Hunger and satiety are controlled by food intake and related hormonal and metabolic adaptations. After a meal, an important signal that has been attributed to drive satiety is glucose, which activates the anorexigenic pro- opiomelanocortin (POMC) neurons in the hypothalamus. Interestingly, only a subpopulation of POMC neurons shows activation by increased levels of glucose. In addition to glucose, lactate, the circulating redox indicator and glucose metabolite, has been shown to affect POMC activity. Indeed, lactate derived either from the circulation or locally by glial cells has been shown to regulate neuronal function, including that of POMC neurons, thus modulating peripheral glucose metabolism as well as feeding. Changes in circulating lactate levels occur in response to feeding, when glucose levels are elevated, and fasting when glucose levels decrease. While administration of lactate has been shown to affect feeding and metabolism via the hypothalamus and POMC neurons, the mechanism(s) via which this process plays a role in physiological control of whole-body energy metabolism in association with glucose is ill-defined. We have recently shown that lactate levels in the circulation and in the cerebrospinal fluid are elevated in fed state and addition of lactate to glucose activates the majority of POMC neurons while increasing cytosolic NADH generation, mitochondrial respiration and extracellular pyruvate levels. Inhibition of lactate dehydrogenases diminishes mitochondrial respiration, NADH production, and POMC neuronal activity. However, inhibition of the mitochondrial pyruvate carrier has no effect. In support of this, MPC deletion in POMC neurons does not alter metabolism. Furthermore, our preliminary metabolic tracing data clearly showed that pyruvate derives predominantly from lactate and that similarly to lactate fatty acids induced increased mitochondrial respiration. Thus, we hypothesize that satiety promotion by POMC neurons require parallel processing of glucose, lactate, and fatty acids. During positive energy balance, glucose in POMC neurons undergo glycolysis thus generating ATP. ATP will then close the KATP channels inducing depolarization and initial activation of POMC neurons. As the ADP:ATP ratio increases, it will drive mitochondrial function which will be supported by lactate oxidation and, thus, generation of NADH to drive the ETC, and by fatty acids oxidation (in addition to lactate) to further drive the TCA cycle. We will determine the role of cytoplasmic (Aim 1) and mitochondrial (Aim 2) redox state in the regulation of POMC neuronal function and associated metabolic adaptations. In Aim 3 we will determine a) the role of fatty acids oxidation in POMC neurons and b) whether glucose, lactate and fatty acids synergistically or uniquely affect POMC neurons. The proposed studies are a logical extension of our preliminary data, and their completion will unmask a fundamental mechanistic principle in the central regulation of metabolism.

NWRS Friends Grant Program

The National Fish and Wildlife Foundation (Foundation) is requesting $100,000 ($87,000 for projects and $13,000 for NFWF operations) to establish a cooperative agreement to support the National Wildlife Refuge System Friends Group grant program. The Foundation and Refuge System have shared the mission and goals of the Refuge System with millions of visitors across the country. This partnership has helped achieve numerous conservation, interpretive, and environmental education and outreach projects and providing additional support will facilitate the continuation of this work through a partnership with a proven track record of success. The National Wildlife Refuge System Friends Group grant program supports start-up, capacity building and specific projects that assist Friends Groups to be effective co-stewards of the important natural resources within the Refuge System. As a special emphasis, a portion of this agreement would be directed towards Friends mentoring projects. The program provides competitive seed grants to creative and innovative proposals that seek to increase the number and effectiveness of organizations assisting the Refuge System nationwide.
