This project will provide recent college graduates with training and increased skill by supporting the fisheries management program at Great Basin National Park (GBNP). This collaborative effort involves the monitoring of fish populations to provide additional information on population status and trends, water quality and quantity, and habitat restoration. Work may include sampling for species presence and distribution, determining water quality and quantity parameters, and assessing and restoring habitat, data entry, data management, quality control and quality assurance. Fisheries management at GBNP focuses on two main issues: reintroduction of the Bonneville Cutthroat Trout (BCT) into its native range and maintaining recreational fishing for the visiting public. The commitment by the NPS to reintroduce BCT at GBNP has helped to forestall its listing under the Endangered Species Act (ESA). The BCT is the only trout native to east central Nevada, and it has experienced major declines caused by natural and anthropogenic changes. This subspecies is endemic to the Bonneville Basin and is a relic of the ancient glacio-pluvial Lake Bonneville. The mission of the Great Basin Institute s Research Associate Program is to provide research opportunities for undergraduate and graduate students, recent graduates, or experienced professionals looking to begin or enhance a career in resource management, conservation, stewardship, and other related fields . The Institute focuses its research efforts on conservation and management of natural and cultural resource in the inner-mountain west, with special emphasis on the Great Basin and Mojave Deserts, as well as the Lake Tahoe Basin. The Research Associate Program provides opportunities related to inventorying rare plant communities, mapping and treating exotic species; conducting recreational use surveys, management of recreation on public lands, wildlife monitoring, cultural resource surveys, and monitoring and evaluating restoration initiatives. Our ecoregion provides substantial opportunities to work in mixed conifer forests, riparian and stream areas, and within a variety of arid land habitats. Research Associate Program project partners include the National Park Service (NPS), as well as numerous other federal, state and county agencies, and NGOs. Personnel from GBI and NPS will collaborate in all aspects of the development of a fish population monitoring and public outreach program. The objective of this project is to participate collaboratively in conservation activities to perform work associated with the monitoring of fish populations in the South Snake Range such as sampling for species presence and distribution ,determining water quality and quantity parameters, and assessing and restoring habitat, data entry, data management, quality control and quality assurance. This project will include fish monitoring in Great Basin National Park and the South Snake range. Travel may be required to visit with experts, attend meetings and symposiums, and to participate in collaborative research, training, and outreach with other agencies such as the Nevada Department of Wildlife (NDOW), the US Forest Service (USFS), and the Bureau of Land Management (BLM).

Natural Families: Engaging Community-Based Organizations to Improve Park Access and Use

The National Park Service, National Capital Region s Rivers, Trails, Conservation Assistance Program (NPS) and the Recipient will create a new and innovative Natural Families plan for engaging community partners and minority families in national parks. The Recipient currently supports more than 250 Family Nature Clubs across the U.S., reaching as many as 50,000 families. Family Nature Clubs are typically structured so that a community leader coordinates family outings in nearby parks and natural areas, exposing families to experiences they might not have had, and places they might not otherwise visit. This significantly adds capacity to the National Park System in its ability to provide innovative approaches to community engagement in parks that leverages relationships with partner organizations. The Family Nature Club model, when propelled in partnership with community-based organizations, maximizes the ability of the National Park System to reach into nontraditional park-goer communities, to build and sustain new relationships with residents in a community, and to bridge generational gaps and empower families to become change agents in their own communities by increasing participation in the national parks in the region.

The purpose of this Notice of Funding Opportunity (NOFO) encourages applications that will expand knowledge of the natural history of disorders that currently are, or may become, part of statewide newborn screening programs. A comprehensive understanding of the natural history of a condition is necessary to facilitate appropriate interventions for infants identified by newborn screening. Characterization of the sequence and timing of symptom development provides information crucial for developing targeted, age-appropriate treatments and for establishing a baseline against which to assess novel interventions. In addition, for some conditions, establishment of genotype-phenotype correlations may facilitate prediction of the clinical course; for others, identification of modifying genetic, epigenetic, or environmental factors may enhance understanding of clinical outcomes. Comprehensive data on the natural history of a condition will facilitate the fields ability to: 1) identify the underlying biological mechanisms; 2) understand the genetic and clinical heterogeneity and phenotypic expression of the condition; 3) improve diagnostic accuracy; 4) facilitate clinical trials by providing comprehensive natural history data; 5) prevent, manage, and treat symptoms and complications of the condition; 6) furnish physicians and families with needed support and predictive information about the condition; and 7) establish data collection systems or patient registries to collect longitudinal data (e.g., child/family outcomes following newborn screening).

Natural Language Processing Phenotyping of Spirituality and Resilience Among Individuals with Opioid Use Disorder

Project Summary Despite vast economic investments to mitigate the harms of opioid use disorder (OUD), the prevalence of the disorder remains high in addition to an estimated 80,000 Americans dying from opioid-related overdoses yearly. Given the significant heterogeneity across multiple dimensions of OUD, prioritizing investigations of multidimensional phenotypes such as resilience may improve our etiological understanding of the disorder. The goal of the proposed study is to develop a laboratory model of resilience, allowing the objective examination of this construct in a controlled laboratory setting. The outpatient study will employ a within-subjects, randomized design and investigate associations of resilience with opioid-seeking and decision-making behaviors. In addition, this study aims to better characterize resilience in individuals with OUD through a deep multidimensional psychological, cognitive, and affective phenotyping approach. To accomplish these goals, individuals who meet DSM-5 criteria for OUD will be recruited to achieve N=125 completers. The primary aim will be to assess whether Resilience Laboratory Tasks (control, cognitive and emotional flexibility) demonstrate convergent validity with self-reported resilience under stress (Maastricht Acute Stress Test, MAST) and non-stress (non-stress test; NST) experimental conditions. Participants will undergo these two experimental sessions (MAST and NST) in random order. During the sessions, heart rate (HR) and HR variability, blood pressure, and galvanic skin response will be assessed as physiological measures of stress reactivity. Immediately following the MAST and NST sessions, participants will complete self-report assessments of stress (perceived stress, craving, and desire to use opioid), a Hypothetical Drug Purchase Task (proxy for opioid self-administration) and Resilience Laboratory Tasks. This proposal will allow the validation of a novel laboratory procedure to empirically examine resilience and provide preliminary data for the psychological, cognitive, emotional, and behavioral substrates of this construct. Additionally, this K08 proposal will provide critical training needed to achieve my career goal of becoming an independently funded scientist-clinician. Specifically, this mentored award will advance my research career by (1) providing essential training in study design and methodology to evaluate the convergent validity of measures of resilience using human stress paradigms, (2) supporting the development of a valid multidimensional phenotypic assessment battery to characterize heterogeneity across multiple dimensions of OUD, (3) improving my skills in advanced statistical methods and allowing me to develop a fundamental understanding of computational modeling, (4) improving my manuscript and grant record. The training goals outlined in this K08 resubmission will equip me with a unique set of skills in human laboratory paradigms and computational modeling which, in combination with the pilot data collected as part of the proposed research, will make me a highly competitive candidate for future R01 submissions.

Natural Products Magnetic Resonance Database (NP-MRD) Phase II

PROJECT SUMMARY/ABSTRACT Since its launch in 2021, the Natural Products Magnetic Resonance Database (NP-MRD, https://np-mrd.org) has become the largest open repository of NMR data on natural products (NPs) as well as the largest collection of general information (nomenclature, structure, biology, chemistry) about NPs in the world. The NP-MRD contains substantial amounts of different types of NMR data including raw NMR data (free induction decays or FIDs), predicted NMR data and literature-derived NMR information (chemical shift assignments, coupling constants). The NMR data in the NP-MRD includes 15,000 sets of experimental NMR spectra (for 3200 NPs), 411,000 simulated NMR spectra (for 21,000 NPs) obtained from literature-derived NMR assignments, and 5.1 million predicted NMR spectra (for all 282,000 NPs). Much of the raw NMR data in the NP-MRD is there due to a concerted effort by NP-MRD, together with the Journal of Natural Products (JNP), to encourage raw data deposition of NMR data used to support novel structure elucidation. For the second phase of the NP-MRD (2025- 2030) we will build on the momentum created in the first phase. Specifically, we will optimize current operations and continue ongoing expansion of both the database content and utility. A major emphasis will be on streamlining NP-MRD operations such that most of its operations will require little human intervention. For example, raw data deposition formerly required two NP-MRD staff in two different locations to interact with the deposited data, and with the depositor. Today, raw data deposition goes automatically into a parser, a set of quality checks and straight into the NP-MRD repository with no human intervention unless (rare) problems occur with the data. Other functions being streamlined, include centralized processes to query, enter and assess NP- MRD data, better processes to annotate NP structures, tools to more efficiently gather biological, medicinal, structural, chemical shift and assignment data from the published literature, as well as workflows to correct entries, remove duplicates and improve accuracy. In terms of data content and utility expansion, major efforts will be undertaken to increase user deposition rates, support the deposition, integration and federation of mass spectrometry (MS) data for NPs into the NP-MRD, implement faster, more “intelligent” NMR, multi-spectral (NMR+MS+CCS+RT) and multi-database (GNPS, MoNA, MassBank, HMDB, FooDB) searches, support more accurate chemical shift, J-coupling and other kinds of “observables” prediction, and create new tools to support automated computer-aided NP structure elucidation (CASE) and dereplication. These streamlining, optimization and database expansion efforts are being done to ensure long-term sustainability of the NP-MRD by making operations cheaper, more automated, more useful and more valuable to the NP community. Outreach to other NP journals and to the NP community will be further expanded, with the expectation that these efforts will triple the data deposition rate to the NP-MRD by 2030, increase community buy-in, increase grant or co-funding support, attract paying subscribers and ensure long-term sustainability of the NP-MRD.
