Lifestyle intervention to improve muscle function in older adults

PROJECT SUMMARY/ABSTRACT In the United States, nearly one third of adults over 60 years of age have sarcopenic obesity which encompasses the combined effect of muscle impairment and obesity. Inducing loss of fat mass while slowing or preventing loss of skeletal muscle mass and quality are critical for preserving the musculoskeletal, cardiometabolic, and cognitive benefits of weight loss in older adults with obesity. As people age, muscle mass and quality decrease while fat mass progressively increases, and these body composition changes are associated with increased incidence of insulin resistance. Calorie restriction and exercise reduce body mass and improve physical performance but lean mass accounts for 22% to 33% of weight loss. Moreover, behavioral adaptations to calorie restriction make adherence a challenge. Energy deficits can be achieved without conscious practice. Since people tend to eat a consistent weight of food, eating a diet low in energy density reduces energy intake. Diets low in energy density reduce body weight and fat mass and moderate the loss of lean mass. Reduced energy intake combined with exercise increases fat loss and improves physical performance more than exercise alone. To date, no adequately powered randomized controlled feeding trial > 14 days has determined the effect of a diet low in energy density on metabolic outcomes. The proposed study will test the hypothesis that a diet low in energy density + exercise (1.0 kcal/g, LD+E) reduces fat mass (Aim 1), improves muscle function (Aim 2) and increases energy deficit (Aim 3). We will conduct a 12-week trial in 80 older adults (> 50 years) with obesity and insulin resistance. Participants will be randomized to LD+E or to the diet typically consumed by older adults + exercise (2.0 kcal/g, TD+E). Baseline energy requirements will be determined using an equation derived from doubly labeled water assessments. All meals for 12 weeks will be prepared in our metabolic kitchen. The macronutrient composition of the diet will be 40-45% carbohydrate; 20- 25% protein; and 35-40% fat. Participants will engage in a combination of supervised aerobic and resistance training for a total of 150 minutes per week, for 12 weeks. Aim 1 will test the hypothesis that LD+E reduces fat mass (dual x-ray absorptiometry) and moderates the loss of lean mass (creatine [methyl-d3] dilution) compared to TD+E. Aim 2 will test the hypothesis that LD+E improves muscle function (short physical performance battery, hand grip strength, six-minute walk test, and VO2 max test). Aim 3 will test the hypothesis that LD+E produces an energy deficit (changes in fat and fat-free mass). Using structural equation models, we will test the effect of circulating concentrations of glucagon-like peptide 1, gastric inhibitory peptide, cortisol, and leptin on energy balance and muscle function. Impairment of skeletal muscle function poses a serious health challenge especially in the context of the demographic trend towards an aging population. The hypothesized outcomes are of far-reaching consequence with the advent of incretin-based pharmacotherapy to treat obesity that causes rapid and significant loss of lean mass comparable to a decade or more of aging.

Light adaptation in regionally and functionally distinct retinal circuits

PROJECT SUMMARY Visual tasks, like reading print or recognizing faces, often require detailed spatial information collected under changing lighting conditions. In the retina, as light intensity varies so do the gain and kinetics of neural responses—a process called adaptation that prevents saturation and supports a consistent perception of contrast. A significant gap in knowledge is how light adaptation functions in the fovea—the central most region of retina responsible for high-acuity vision. Retinal circuit adaptation relies on signal pooling, and therefore adaptation may vary between foveal and peripheral regions that differ in convergence. Additionally, cone photoreceptors in peripheral primate retina are known to be asymmetric in their sensitivity to light increments vs decrements. Therefore, circuit adaptation may differ in ON and OFF visual pathways. The objective of this project is to use distinct primate retinal circuits—foveal vs peripheral and ON vs OFF—to determine how circuits with differing signal to noise ratios modulate gain and kinetics across light conditions. Aim 1 will determine the impact of convergence on properties of retinal circuit adaptation (gain, kinetics, noise, and time course) in the primate foveal vs peripheral midget and parasol pathways, as well as the contribution of synaptic inhibition as a potential mechanism for circuit adaptation to luminance. Aim 2 will determine how asymmetries inherited from cone photoreceptors shape the functional properties of adaptation in the primate ON vs OFF peripheral midget pathway. Given the importance of the fovea for our everyday vision, this work will bridge a gap in our knowledge of how foveal circuits adapt to changes in contrast over varying background luminance, critical for the function of high-acuity vision. These results will positively impact the pursuit towards prosthetic retinal implants that can recapitulate properties of foveal circuits by providing a template for function in diverse retinal circuits. The training plan described in this proposal is designed to enable me to develop the skills necessary to reach my career goal of an independent investigator. By following this plan with the guidance of my sponsor and co-sponsor, I will continue to learn new electrophysiology techniques to build a strong foundation in retinal circuit research. I will develop my writing, communication, teaching, mentoring, networking, and scientific outreach skills. This research will take place at the University of Wisconsin-Madison where the strong intellectual environment and availability of primate tissue from the Wisconsin National Primate Research Center will be leveraged.

Light-Assisted Drying: A Novel Method for Stabilizing Inactivated Poliovirus Vaccine in a Sugar Glass for Room-Temperature Storage

PROJECT SUMMARY Cold chain logistics are critical for maintaining the stability of biological products, including vaccines, monoclonal antibodies, protein-based therapeutics, and diagnostic specimens. However, the reliance on continuous refrigeration poses substantial logistical and financial challenges, particularly in low-resource settings. Developing alternative stabilization methods that eliminate refrigeration requirements is crucial for improving global access and reducing costs. Current strategies, such as engineering inherently stable biomolecules, are labor-intensive and complex. Conventional techniques, including freeze, vacuum, spray, and thin-film drying, expose biologics to extreme temperatures and/or pressures, require long processing times, and depend on complex formulations. Thus, a more efficient, scalable, and cost-effective stabilization technology is needed to preserve biological activity while enabling ambient-temperature storage. We have developed a novel technique, light-assisted drying (LAD), to create trehalose-based amorphous solids for biologic preservation. Using a near-IR laser, LAD rapidly removes water from a liquid formulation, forming a protective trehalose matrix that stabilizes the biologic. By precisely controlling sample temperature, LAD prevents damage to thermally sensitive biologics. In contrast to traditional methods, LAD processes samples quickly and requires minimal excipients, while avoiding exposure to extreme temperatures or pressures. We have successfully applied LAD to the model protein lysozyme and nucleic acid nanoparticles used in nanomedicine. Additionally, we have demonstrated LAD’s capability to process volumes ranging from 0.25 to 1 mL—typical vaccine doses—and to process samples inside glass vials commonly used in industry. We have also developed a system capable of processing multiple samples simultaneously, highlighting LAD’s scalability and versatility for biologic preservation. This study will evaluate LAD’s ability to stabilize inactivated poliovirus vaccine (IPV). Our preliminary data suggest that LAD preserves IPV potency. We will build on these findings by assessing humoral immune responses in a mouse model following immunization with the LAD-processed vaccine. Long-term stability will be evaluated through storage trials at both freezing and elevated temperatures. Potency will be measured using ELISA assays, and immunogenicity for stored vaccine will be examined via in vivo testing. Additionally, transmission electron microscopy (TEM) will assess the structural integrity of the viral capsid both immediately post-processing and after storage. This project will validate LAD for IPV stabilization, marking the first application of the technology to a vaccine, and establish LAD as a versatile platform for preserving other biologics. The findings will provide essential data for broader applications of LAD and future translation and commercialization efforts.

Light-Responsive Inorganic Compounds for Diagnosis and Therapy

Surgery, alone or in combination with other therapies, is a cornerstone of cancer treatment. However, incomplete resection leads to increased risk of recurrence and reduced overall survival. Here optical image guided surgery (IGS) has a role to play by enhancing tumor visualization and improving surgical precision. It has notable advantages over visual inspection and histology aided by real-time CT, MRI, or optical scanners alone, but could be improved with new optical imaging agents. IGS could benefit from imaging agents that luminesce brightly in the wavelength region where tissue is most transparent. Further, IGS agents that also exert potent antitumor effects would enable simultaneous detection and treatment of disseminated cancer cells that cannot be removed due to their location around critical organ structures. This project will design osmium compounds that are highly luminescent at wavelengths where tissue is the most transparent. The optical agents will be inert and nontoxic, but phototoxic toward cancer cells through complementary modes of action, regardless of oxygenation status. The multidisciplinary strategy is based on the unique photophysical properties of certain immunostimulatory osmium compounds, with specialized ligands, that can be independently tuned for near-infrared luminescence output and phototherapy with minimal impact by oxygen. The molecular properties of these inorganic compounds can be further augmented by encapsulation in supramolecular structures such as nanolipid carriers. The project scope is to synthesize and characterize new compounds, determine their molecular properties, interrogate their photophysical and cellular mechanisms of action, and evaluate their utility as multifunctional optical imaging agents with therapeutic capacity. These activities will expose undergraduate students to multidisciplinary research and create new knowledge regarding optical imaging agents that addresses current challenges that have so far prevented IGS from being effectively used for hard-to-resect cancers. This project will also strengthen the research environment at UTA by enabling long-term sustainable biomedical research and inspiring a new generation of students to pursue innovative solutions to critical human health challenges.
