Mitigation is the effort to reduce loss of life and property by lessening the impact of disasters. Mitigation is taking action now, before the next disaster, to reduce human and financial consequences later. Effective mitigation requires that we all understand local risks, address the hard choices and invest in long-term community well-being and resilience. Without mitigation actions, we jeopardize our safety, financial security, and self-reliance. As the result of a Presidential Disaster Declaration, FEMA’s Hazard Mitigation Grant Program (HMGP) funds plans and projects that reduce the effects of future natural disasters. In California, these funds are administered by the Cal OES HMGP Unit. Eligible subapplicants include state agencies, local governments, special districts, and some private non-profits. The NOI is intended to provide an opportunity for eligible subapplicants to propose well-defined mitigation actions that reduce risk to life and property from future natural hazards. Eligible subapplications that are not initially selected for submission to FEMA will be retained for future consideration when funding becomes available. Eligible Subapplicants include state agencies, local governments, special districts, and Federally-recognized tribes. Sub-applicants must have a FEMA-approved and locally adopted Local Hazard Mitigation Plan (LHMP) to be eligible for project grants. If your agency or jurisdiction does not have a LHMP, you may apply for grant funding to develop one. Single jurisdiction plans are limited to a maximum federal share of $150,000; Multi-jurisdiction plans are limited to a maximum federal share of $250,000. Some private nonprofit organizations are eligible for HMGP funding, consistent with 44 CFR §206.221and 206.434. Private nonprofit organizations are not required to have an LHMP to be eligible, but the County they are located in must meet the LHMP requirement.

HCN Channels Mediate Olfactory Dysfunction in Aging and Neurological Disease

Project Summary/Abstract Olfaction frequently declines during aging and in neurological diseases including Alzheimer’s disease. Hyperpolarization- activated cyclic nucleotide-gated cation (HCN) channels (isoforms 1-4) generate the Ih conductance, playing controlling roles in neuronal excitability and rhythmic firing activity in individual and networks of neurons. HCN channel dysfunction is commonly observed in aging and almost all neurological diseases. This proposal uncovers a novel mechanism of olfactory bulb function and dysfunction in which there is aging/disease related decline in HCN channel regulation of excitatory neuron output from mitral/tufted cells (M/TCs) in olfactory bulb. Large Ih currents are present in external tufted cells (eTCs, a class of M/TCs), acting as a pacemaker current generating spontaneous intrinsic bursts, which drive long-last depolarizations and cluster firing of deeper output neurons. Our overarching hypothesis is downregulation of HCN channels during aging and Alzheimer’s disease disrupts output neuron firing leading to diminished performance on olfactory mediated behavioral tasks. Across four aims we will undertake a project spanning from the gene to cells, to circuits, and to behavior, linking HCN channel expression to olfactory bulb output neural activity, to olfactory performance in health and aged/disease. We will show that there is a significant and selective reduction in HCN4 channel expression and Ih conductance during aging and in an animal model that recapitulates some elements of familial Alzheimer’s disease (the 5xFAD mice, Aim 1). Furthermore, we will demonstrate that expression decline correlates with impaired Ih mediated activity of olfactory bulb output neurons in aging/disease and that pharmacological suppression of Ih in young mice recapitulates aspects of aging/disease olfactory network activity disruption (Aim 2). In Aim 3, we will show pharmacological manipulation of HCN channel function in the olfactory bulb causes diminished behavioral performance in young animals on olfactory mediated tasks, similar to aging/disease related performance decline. Finally, in Aim 4 we will manipulate HCN4 expression selectively in olfactory bulb output neurons (using a vGlut2-cre line of mice with viral knockdown construct) to show that downregulation of HCN4 in young olfactory bulbs ‘ages/diseases’ the network, causing output neuron activity and behavior to diminish to that of an aged/diseased olfactory bulb. Furthermore, we will show that olfactory decline in aging/disease can be ‘rescued’ by HCN4 upregulation selectively in olfactory bulb output neurons (vGlut2-cre line of mice with a viral overexpression construct). Upregulated HCN4 restores Ih currents, increases M/TC neural activity, and ameliorates olfactory behavior deficits. What we show in robust preliminary data is that pharmacological and genetic manipulation of olfactory bulb output neuron HCN4 expression can artificially ‘age’ younger animals or ‘rejuvenate’ an aged/diseased mouse to rescue performance. Taken together, these aims are a coordinated investigation of HCN channel function/dysfunction in the excitatory cell network of the olfactory bulb in adult, normal aging, and an animal model of neurodegenerative disease. The results of this study highlight new circuit mechanisms and potential therapeutic targets for mitigating olfactory deficits.

New advances in imaging, biomechanics, tissue-specific omics, electrophysiology, and digital health tools now allow for a more complete evaluation of the whole joint, including periarticular tissues such as muscle, adipose, ligaments, tendons, and fascia.The phased projects supported by this program will first identify and validate multi-tissue mechanisms underlying joint pain, then test non-pharmacological or multimodal interventions that directly target these mechanisms. Please note that while applications can specify the length of time for each of the following study phases, the total project length cannot exceed 5 years:Phase 1 (2 to 3 years): Projects will identify multisystem mechanisms that include signals from periarticular tissues (e.g., muscles, adipose, and connective tissues including ligaments, tendons, or fascia) and their potential interactions with articular tissues (e.g., bone, synovium, cartilage) or the peripheral nervous system that contribute to joint pain and pathophysiology. Effective management of both conditions is hindered by siloed medical disciplines and health systems. HEAL Integrative Management of Chronic Pain and OUD for Whole Recovery (IMPOWR) was funded in FY21 to support 11 unique clinical trials to test novel interventions for the management of co-occurring chronic pain and OUD at the patient-level. The current NOFO will develop strategies to address barriers to the sustainable and effective delivery of integrated OUD/pain care at the health systems level. IMPOWR identified patient interventions that were effective and ready to implement. The current NOFO will address how health systems could be improved to facilitate the use of effective integrative interventions, including the involvement and roles of peer specialists, clinicians, health system leaders and other key decision makers. Also, this NOFO will characterize where such care can be optimally and efficiently delivered. Studies will identify collaborative care models, implementation strategies, and other innovative health system approaches to meaningfully integrate CP and OUD service provision in health settings. These studies will address complex factors that influence the ability to scale and sustain effective collaborative and integrated care for pain and OUD. The National Institute on Drug Abuse (NIDA) in partnership with National Institute on Alcohol and Alcoholism (NIAAA), National Institute of Arthritis and Musculoskeletal and Skin Diseases (NIAMS), National Institute of Mental Health (NIMH), and National Center for Complementary and Integrative Health (NCCIH), and National Institute of Neurological Disorders and Stroke (NINDS) intends to publish a Notice of Funding Opportunity (NOFO) to solicit applications for research on the effective management of OUD and CP. Studies will execute a single hybrid II/III implementation-effectiveness trial to focus on collaborative care models and implementation strategies to meaningfully integrate CP and OUD service provision in diverse health settings. These studies will address complex factors that influence the ability to scale and sustain effective and integrated care for CP and OUD. These studies will address complex factors that influence the ability to scale and sustain effective and integrated care for CP and OUD. The BEACON Center will provide: (1) network coordination support, (2) capacity building at the intersection of CP, OUD, and implementation science, (3) annual surveys/qualitative activities to understand different barriers and attitudes impacting access and sustained use to integrated CP and OUD service delivery, (4) develop important resources for key stakeholder audiences that can inform/improve sustainability and scalability of evidence-based practices.The National Institute on Drug Abuse (NIDA) in partnership with National Institute on Alcohol and Alcoholism (NIAAA), National Institute of Arthritis and Musculoskeletal and Skin Diseases (NIAMS), National Institute of Mental Health (NIMH), and National Center for Complementary and Integrative Health (NCCIH), and National Institute of Neurological Disorders and Stroke (NINDS) intends to publish a Notice of Funding Opportunity (NOFO) to solicit applications for research on the effective management of OUD and CP. Applications are not being solicited at this time. Notice is being provided to allow potential applicants sufficient time to develop meaningful collaborations and responsive projects. This NOFO will utilize the U2C activity code. Investigators with expertise and insights into this area of CP management and OUD/opioid misuse are encouraged to begin to consider applying for this new NOFO. In addition, collaborative investigations combining expertise in community-based participatory research approaches, implementation science, and other strategies to amplify scalability and sustainability will be encouraged and, these investigators should also begin considering applying for this NOFO. This Center will support the coordination and integration of data from both the HEAL INTERACT program and progressively facilitate broader integration of related musculoskeletal datasets and programs. The Data Coordination and Integration Center will lead efforts for managing and integrating complex pain-associated data types and information from the INTERACT Consortium and progressively related datasets. This is a Notice of Funding Opportunity (NOFO) is for a Limited Competition that will invite applications from eligible organizations to apply. Please see Section III. Eligibility, for additional information. In accordance with NIH standard peer-review processes, the applications will be peer-reviewed, and only meritorious applications will be considered for funding. This NOFO will renew the HEALthy Brain and Child Development (HBCD) Consortium Administrative Core (HCAC) and Data Coordinating Center (HDCC).The HBCD Study is a nationwide, multi-site, longitudinal study that follows children from birth through childhood. The study examines brain and behavioral development, with a focus on how prenatal substance exposure affects health outcomes.The HBCD Consortium consists of three highly integrated components: (1) a set of linked Research Project Sites, (2) a single overall HCAC; and (3) a single central HDCC. As such, this NOFO runs in parallel with a companion NOFO that solicits applications for linked research sites (RFA-DA-27-013). It is expected that investigators, upon funding, will work jointly with NIH scientific staff to assist, guide, coordinate, or participate in project activities.
