About This Grant
Project summary The overall goal of this application is to uncover the effects of Tannerella forsythia (Tf) secreted methylglyoxal on periodontal inflammation and microbial dysbiosis. Tf is an understudied Gram- negative anaerobe implicated in the development of periodontitis. This bacterium has a unique ability to produce methylglyoxal (MGO), an electrophilic and highly reactive dicarbonyl compound that can covalently modify proteins to generate immunogenic glycated adducts known as the advanced glycation endproducts (AGEs). These AGEs can promote inflammation by activating the receptor for the AGEs (RAGE). Moreover, MGO can disrupt host and bacterial protein structural and functional integrity by forming their glycation adducts, contributing to inflammation and dysbiosis via other pathways. A possible role of MGO secreted by Tf is supported by previous studies indicating that the disease severity characterized by increasing clinical attachment loss (pocket depth) correlates positively with the MGO concentrations in the gingival crevicular fluid (GCF) and Tf bacterial load. The involvement of this bacterial -produced MGO in inflammation is supported by our data demonstrating that Tf-MGO by modifying host proteins, such as extracellular matrix collagen into AGE adducts, can induce RAGE-dependent activation of inflammatory cytokine secretion in human monocytic cells and human gingival fibroblasts (HGFs). In addition, Tf-secreted MGO disrupts the biogenesis and structural integrity of fimbriae produced by the cohabiting key stone oral pathogen P. gingivalis. The role of bacterially produced MGO and downstream glycation adducts formed by MGO in periodontal inflammation and dysbiosis has never been explored before. In this application we will explore the role of T. forsythia produced MGO in periodontal inflammation and dysbiosis via two specific aims. Aim 1. To identify and determine the impact of T. forsythia-MGO modified host and bacterial proteins on inflammation and bacterial virulence. Specific Aim 2. To investigate the role of T. forsythia-infection induced AGEs on periodontal inflammation and dysbiosis. The expected outcomes of this proposal will shed new light on the underlying mechanisms of disease pathogenesis induced by MGO and glycated adducts formed by Tannerella forsythia. This research could potentially lay the groundwork for designing novel therapeutic approaches to target T. forsythia-Methylglyoxal-AGE-RAGE-axis to treat periodontitis.
