Pol III activity is, itself, directly linked with modulating self-renewal and differentiation, and Pol III- derived ncRNAs individually promote cell proliferation and migration phenotypes via RNA-based mechanisms, altogether linking Pol III transcription dynamics and downstream small RNA landscapes to changes in cellular physiology. However, the full breadth of Pol III transcription and the mechanisms that shape tissue-specific patterns remain gaps in fundamental knowledge due to limited holistic studies of Pol III activity in cells. This proposal leverages a unique combination of genomic and biochemical strategies to systematically profile the breadth of Pol III activity and uncover new regulatory mechanisms - focusing first on a disease-associated Pol III subunit (RPC7a) and the Nucleosome Remodeling and Deacetylase (NuRD) complex. This cross-modality framework applies innovative genomic techniques, such as direct Pol III-4SU-UV-CLIP-nascent-seq and orthogonal analysis of Pol III-centered RNA chaperone protein La (SSB) ncRNA interactions, as well as traditional in vitro transcription tests to characterize both cellular and biochemical mechanisms. Determining the full range of Pol III transcription and the mechanisms contributing to context-specific Pol III regulation will have direct relevance to understanding Pol III dysregulation in cancer, infection, and other diseases, and will open new avenues for research on basic mechanisms of transcription regulation.
