Project Summary The concept of health and disease has shifted with the growing recognition that animals, including humans, are holobionts—integrated systems of hosts and microbial symbionts. While most animals harbor multiple microbiomes across tissues, the systemic regulation of these communities remains poorly understood. The Hawaiian bobtail squid, Euprymna scolopes, is a model organism used to study the influences of beneficial bacteria on animal development, interactions with mucosal epithelia, and the innate immune system. The host has two well-characterized microbiomes, the light organ with a single symbiont, the bioluminescent bacterium Vibrio fischeri, and the accessory nidamental gland, a female reproductive organ with a bacterial consortium involved with producing antimicrobials involved with egg defense. The focus of this research will cover several themes: 1) understanding systemic regulation of different microbiomes via the innate immune system; 2) influence of different microbiomes on blood cell development (hematopoiesis) and development of mucosal epithelia and organogenesis; and 3) implementation of gene editing methods in the squid-bacteria system to study host-microbe interactions under controlled gnotobiotic conditions. Overall, this work will determine mechanisms by which animals regulate different microbiotas, filling a major gap in our understanding of host-microbe associations that have direct relevance to human health and disease.
