About This Grant

ABSTRACT Zebrafish (Danio rerio) are a critical model organism in genetic and biomedical research due to their genetic similarity to humans, rapid embryonic development, and utility in studying a wide range of diseases. Researchers have over 45,000 zebrafish lines, but maintaining these genetic resources is costly, logistically challenging, and prone to genetic drift, particularly for lines with complex genetics and/or mitochondrial and maternal traits that cannot be preserved via sperm cryopreservation. Reliable zebrafish embryo cryopreservation methods would resolve these challenges, enabling long-term storage and recovery of entire (diploid and mitochondrial) genomes. Our group has developed the first successful zebrafish embryo cryopreservation protocol, combining microinjection of cryoprotectants (CPAs) and plasmonic nanoparticles with laser rewarming. This method has demonstrated survival to adulthood with normal fecundity and physiology, with recent refinements achieving near 10% survival rate to day 5 post-fertilization. To accelerate the cryopreservation and archiving of critical zebrafish stocks, we will develop, standardize, and disseminate resources to individual labs and stock centers. Significant efforts will be devoted to developing reliable methods for cryogenic storage and shipping of zebrafish embryos as well as to study if any significant mutagenic changes occur due to cryopreservation. We will be working with multiple groups within the cryobiology and zebrafish community to gather feedback on and refine our approach. With our collaboration with the Zebrafish International Resource Center (ZIRC), we will identify and prioritize the stocks that need cryopreservation to prevent their total loss in the event of a disaster or genetic drift. We will also demonstrate the efficacy of our cryopreservation protocol on a large scale with a selected group of wild-type and mitochondrial mutant lines identified by the Ekker Lab at UT Austin. Finally, we will also estimate the costs for cryopreservation in comparison to the traditional method of zebrafish maintenance to inform future efforts to protect these precious genetic resources. By reducing the financial and logistical burden of maintaining live zebrafish lines, this project will streamline genetic resource management, preserve complex genetic traits, and enhance global accessibility and collaboration in zebrafish research. This is a multi-PI project led by investigators with significant experience in zebrafish and cryobiology.
