About This Grant
ABSTRACT HIV infection and anti-retroviral therapy (ART) are associated with multiple chronic metabolic complications that occur more frequently and at earlier onset than people without HIV, including obesity, cardiovascular disease, chronic kidney disease, osteoporosis, cancers, frailty, and cognitive decline that significantly diminish their quality of life. HIV and its treatments likely contribute to these metabolic problems. However, the reasons for metabolic diseases in PLWH remain unknown. A significant barrier to understanding chronic conditions among PLWH is the complete lack of models that simulate the disease seen in people. This application directly responds to the Funding Opportunity Announcement PAR-25-068 which encourages cellular and molecular characterization of the HIV reservoirs in adipose tissue. Our project will develop preclinical mouse models that express one protein of HIV that is known to cause phenotypes resembling the condition in PLWH. We will combine knowledge learned from mouse experiments to understand organ-specific changes that provoke the chronic conditions that lead to longer-term co-morbid conditions seen in PLWH. There is an urgent need to understand the mechanisms that cause heterogeneous metabolic disorders among the many PLWH living normal life spans in the post-ART era. Our proposed research is designed to address these urgent needs and provide vital tools for the field-at-large.
