About This Grant

ABSTRACT Atherosclerosis, a primary driver of cardiovascular disease, results from the accumulation of lipids and immune dysregulation in arterial walls. Current therapies, such as statins, are limited in their ability to address immune-mediated mechanisms. Human genetic studies identify CXCL5 as a causal gene for coronary artery disease, with lower levels linked to severe disease. Our preliminary data show that CXCL5 enhances macrophage cholesterol efflux and prevents plaque progression in mouse models. This multi-PI proposal integrates systems biology, pharmacology, and atherosclerosis expertise to elucidate CXCL5- CXCR2 signaling mechanisms using multi-omics and CRISPR-Cas9 in human macrophages (Aim 1), maps CXCL5-dependent immune responses via single-cell RNA sequencing in mouse models and human data (Aim 2), and evaluates CXCL5’s therapeutic potential to prevent and reverse atherosclerosis in preclinical models (Aim 3), aiming to develop novel immune-based therapies for cardiovascular disease management.
