About This Grant

Project Summary Glaucoma is a leading cause of blindness affecting 80 million people worldwide. The mechanisms underlying glaucoma have been studied extensively but are not yet fully understood. Our lab has a long-term interest in ocular lymphatic research. This project is based on our recent discovery of a new aqueous humor outflow pathway that drains from the Schlemm’s canal to conjunctival lymphatic vessels. This new lymphatic outflow pathway is distinctive from the conventional pathway where aqueous humor drains from the Schlemm’s canal to episcleral veins, which are blood vessels. This novel discovery offers a new concept to study ocular fluid drainage and the pathogenesis and treatment of ocular hypertension and glaucoma. The goal of this proposal is to further investigate this novel discovery and to develop new strategies to treat ocular hypertension and glaucoma. Our hypothesis is that the conjunctival lymphatic outflow pathway plays an important role in aqueous humor drainage. Its modulation can lower intraocular pressure and treat glaucoma. This project is based on a large amount of preliminary data and the most recent advances in science and technology. Our specific aims are to 1) assess the specific role of the conjunctival lymphatic pathway in aqueous humor drainage and IOP regulation; 2) investigate the effects of pro-lymphatic treatment on the conjunctival lymphatic pathway; and 3) explore the effects of pro-lymphatic treatment on ocular hypertension and glaucoma. Our study promises to reveal novel mechanisms and therapeutic strategies for glaucoma, and possibly other lymphatic-related diseases in the body.
